"SYNTHESIS AND SPECTRAL STUDIES OF ESTER DERIVATIVES OF NAPROXEN"

ABSTRACT
Naproxen and Naproxen sodium are Non-steroidal anti-inflammatory drugs that inhibit prostaglandin synthesis.Naproxen is a derivative of propionic acid.Naproxen is clinically effective in patients with postoperative pain.This NSAID is used for the treatment of mild to moderate pain,inflammation,fever,rheumatoid arthritis,osteoarthritis,juvenile forms of arthritis, acute gout,dysmenorrhea,migraine,headache,premenstrual syndrome and ankylosing spondylitis.
The gastrointestinal ulcerogenicity posses major drawback in the use of Naproxen as NSAID for long term therapy of arthritis and other rheumatic diseases. There is always a need to safer NSAID and research efforts are on going for developing safer NSAID’S.The prodrug approach is one of the most promising ones in these approaches. The most common approach for preparing a prodrug is to change the drug containing -COOH or -OH group to esters; so the prodrug of anti-inflammatory agents liberate the active species upon metabolism. Many ester derivatives of Naproxen have been prepared and it found effective. Ester prodrugs have been proposed to be potentially useful prodrug candidates for NSAID’S.
Ethyl-2-(6-Methoxy napthyl) propionate and Iso-propy-2-(6-Methoxy napthyl) propionate can be prepared as a ester prodrug by converting -COOH group of Naproxen to -COOR group in presence of sulphuric acid and respective alcohol moiety.
