Quantitation of Rutin in Methanolic Leaf Extract of Phyllanthus reticulates Poir. by High Performance Thin Lay
er Chromatography

Abstract

    A sensitive, simple and accurate High Performance Thin Layer Chromatographic method has been established for determination of rutin in leaf powder of Phyllanthus reticulates Poir. A Methanolic extract of leaf powder was used for experimental work. The concentration of rutin in leaf was found to be 2.503% w/w. Separation was performed on aluminum backed silica gel 60 F254 HPTLC plates with ethyl acetate-butanol-formic acid-water (50:30:10:10 v/v) as mobile phase. After development plates were dried in oven at 100 șC for 5 min. Detection and quantification were performed by densitometric scanning at 366 nm. The method was validated for its precision and accuracy. The accuracy of the method was checked by conducting recovery studies at three different levels of rutin and the average % recovery of rutin was found to be 97.71%. The proposed HPTLC method provided good resolution of rutin from other constituent present in methanolic extract of dry leaf powder of Phyllanthus reticulates Poir. and can be used for quantification of rutin present in the leaf extract or marketed formulation of Phyllanthus reticulates Poir.
