PREPARATION AND CHARACTERIZATION OF SPHERICAL CRYSTALS OF VALSARTAN FOR DIRECT COMPRESSION METHOD

Abstract
Valsartan is an anti-hypertensive drug, characterized by its poor flow property, aqueous solubility, wettability and compressibility. The main objective of present study was to prepared spherical crystals of valsartan and to study effect of spherical agglomeration on micromeritics properties, wettability, compressibility and aqueous solubility.
Valsartan drug were modified into spherical crystals using Acetone-Chloroform-Water as solvent system into ratio 5: 0.75: 50 respectively. Spherical agglomerates were characterized by IR Spectroscopy. Flowability was determined in terms of Angle of repose, Carl’s consolidation index, Compressibility index and Hausner’s ratio. Compressibility was determined using Hardness tester. Wettability was determined by calculating contact angle. Aqueous solubility was determined by preparing saturated aqueous solution of drug and Spherical crystals of valsartan.. Spherical crystals of valsartan showed improved micromeritics properties and compression properties, wettability, aqueous solubility( nearly 10 times) in comparison to conventional drug. So spherical crystals of valsartan can be used for direct compression method with improved aqueous solubility. IR Spectra indicates no drug: solvent interaction occurred in spherical crystals of valsartan.
